PRODUCT DESCRIPTION INTENDEND USE Siron is an activated partial thromboplastin time (aPTT) reagent to be used: - as a screening test for the intrinsic coagulation system - in the diagnosis and treatment of haemophilia A and B - for specific determinations of factors VIII, IX, X, XI and XII* when used in combination with the corresponding deficiency plasma. for control of heparin treatment - for detection of Lupus anticoagulant Siron measures the overall activity of the intrinsic coagulation system. Siron is sensitive to the factors XII, XI, X, IX, VIII, V and II, but insensitive to factors VII and XIII. * For FXII clotting assays a 1:20 dilutions should be used. COMPOSITION Siron is an activated partial thromboplastin time (aPTT) test. This liquid preparation is a stable aqueous suspension of phospholipids. FXII activation of Siron is achieved by addition ellagic acide. STABILITY AND STORAGE The expiry date printed on the labels applies to storage of the unopened bottles at +2…8 °C. MANUAL The CaCI2 25 mmol/L should be preheated to 37°C. Pipetting Scheme: 100 µL plasma sample + 100 µL Siron in a prewarmed tube incubate for 4 minutes + 100 µL CaCI2 solution (25 mmol/L;37°C) Start stop-watch simultaneously Leave the tube for 20 sec. before tilting. Determine the clotting end point when clot is formed. The results are given in seconds. REFERENCE RANGE Normal range: 25-45 sec. The normal range refers to manual method. It is recommended that each laboratory establish and controls its normal range.
